Optimized in Vitro Pigmentation Screening Assay Using a Reconstructed Three Dimensional Human Skin Model

چکیده
Abnormal pigmentation is related to a variety of cosmetic and clinical conditions including melasma, lentigo, age spots, vitiligo, etc. Numerous actives designed to modulate skin tone have been utilized as cosmetics and pharmaceuticals to address these conditions. We report on an optimized short-term (1-week) protocol using the Asian MelanoDermTM model (MatTek Corporation) and designed to rapidly evaluate candidate actives that modulate melanin production in human skin. Initial assay development activities identified the shortest in-life assay phase sufficiently sensitive to identify melanogenesis modulators. Based upon the initial results, an optimized protocol was subsequently evaluated for reproducibility in four independent trials.

Development of melanocye-keratinocyte co-culture model for controls and vitiligo to assess regulators of pigmentation and melanocytes.
BACKGROUND There is a need to develop an in vitro skin models which can be used as alternative system for research and testing pharmacological products in place of laboratory animals. Therefore to study the biology and pathophysiology of pigmentation and vitiligo, reliable in vitro skin pigmentation models are required. AIM In this study, we used primary cultured melanocytes and keratinocytes...
متن کاملGenotoxicity assays with Episkin, a reconstructed skin model: Towards new tools for in vitro risk assessment of dermally applied compounds?
In vitro reconstructed human skin such as Episkin® is a widely used model in safety or efficacy pre-screening tests. It is of growing interest for regulatory purposes as an alternative to animal testing. The reduction and eventually the replacement of In vivo toxicity testing require the development of In vitro models to predict the genotoxic or other endpoint risk. This can be achieved if thes...
متن کاملSolar ultraviolet radiation induces biological alterations in human skin in vitro: relevance of a well-balanced UVA/UVB protection.
Cutaneous damages such as sunburn, pigmentation, and photoaging are known to be induced by acute as well as repetitive sun exposure. Not only for basic research, but also for the design of the most efficient photoprotection, it is crucial to understand and identify the early biological events occurring after ultraviolet (UV) exposure. Reconstructed human skin models provide excellent and reliab...
متن کاملKey Regulatory Role of Dermal Fibroblasts in Pigmentation as Demonstrated Using a Reconstructed Skin Model: Impact of Photo-Aging
To study cutaneous pigmentation in a physiological context, we have previously developed a functional pigmented reconstructed skin model composed of a melanocyte-containing epidermis grown on a dermal equivalent comprising living fibroblasts. The present studies, using the same model, aimed to demonstrate that dermal fibroblasts influence skin pigmentation up to the macroscopic level. The proof...
متن کاملI-5: Multicellular Human Testicular Organoid: A Novel 3D In Vitro Germ Cell and Testicular Toxicity Model
Background Background: Mammalian spermatogenesis is regulated through paracrine and endocrine activity, specific cell signaling, and local control mechanisms. These highly specific signaling interactions are effectively absent upon placing testicular cells into two-dimensional primary culture. The specific changes that occur between key cell types and involved spermatogenesis signaling pathways...
